Immunopharmacology Comments
TLR6 forms heterodimers with TLR2 to detect a wide range of bacterial lipopeptides (LP) from bacterial cell membranes, and effect an immune response [1]. TLR2/6 dimers perferentially detect diacylated LP [2]. Pam2Cys-SKKKK is a synthetic LP which induces signalling through TLR2/TLR6 heteromers. Activation of TLR2-TLR6 heteromers by LP promotes formation of a functional signalling complex that includes MyD88 and IL-1R-associated kinase (IRAK) proteins, that ultimately initiates NF-κB-mediated cytokine production.
